MC3R and MC4R are class A G protein-coupled receptors coupled through Gs to adenylyl cyclase. Agonist binding raises intracellular cAMP and activates protein kinase A, with downstream CREB phosphorylation. MC4R in particular is densely expressed in the paraventricular nucleus of the hypothalamus and in limbic structures, and its signalling is antagonised endogenously by agouti-related peptide, which gives laboratory models a defined counterparty for competitive binding work.
